Since The Joker movies is a hot topic at the moment, I thought I would link the comicbook it takes elements from.

Batman: The Killing Joke is a 1988 comicbook written by Alan Moore and is considered to be the definitive Joker story and one of the best Batman stories ever published.

It begins with Batman realising that his feud with the Joker will only end with one of them killing the other, and he goes to Arkham Aslyum to try and talk the Joker down once and for all, only to discover that the Joker has escaped.

The Joker sets out to prove a point to Batman that it only takes "one bad day" for anyone to become like himself. He does this by shooting Barbara Gordon (Batgirl) through the spine paralysing her for life, and then sending her father Jim Gordon through a ghost train ride where he is forced to view pictures of his naked daughter as the Joker psychology tortures him.

The Joker's origin is presented via flashback where it is revealed that he was once a loser and a struggling comedian with a pregnant wife. He takes a one off job with a couple of criminals to pull off a robbery, but after tragedy strikes it leads to his transformation as the Joker.

However given the ambiguous nature of the Joker, this origin story is left up in the air with the Joker later remarking in the story that some days he barely remembers the events that tipped him over the edge and that when it comes to his past he prefers it to be multiple choice.

There is a theory amongst some fans that The Killing Joke ends with Batman finally crossing the line and killing the Joker. In the last panels of the book they are seen holding onto each and laughing hysterically at a joke the Joker makes, when suddenly the Joker's laugh abruptly stops leaving Batman laughing alone for a couple of panels and then silence. The implication is that Batman strangles the Joker to death.

Of course if true this is not the first time the Joker has died in the comics, in fact he was originally intended to be a one-off character when he first appeared in Batman (April 25, 1940). At the end of that story he was supposed to die after accidentally stabbing himself in a fight with Batman. However the editor of the comic saw something in the Joker and thought he would make a great recurring villain, and the ending was rewritten with the police finding the Joker still breathing with the knife in his chest.

The best Joker death ever however goes to Superman in Injustice (Year One, Part 1)...
